What do you use to seal these?

They hold two coins each and fold in such a way to keep the coins from falling out so you don't need to seal them. These are inert and safe for very long term storage so I wouldn't use tape or anything sticky because you don't know what chemicals are in the sticky.

Which is the best size for the American Silver Eagle, 2" or 2.5"?

Either will work. American Silver Eagle uses a 40.6 mm (1.6") direct fit capsule and the SAFLIP is 2" wide but you must account for depth also. 2" will be a tight fit. Personally, I suggest the 2.5" for most 40mm coins because they average about 3mm depth.
